The present invention is directed to a method for localizing, in chromatin or DNA, the binding loci of a chromatin or DNA binding protein comprising the following steps:a) tethering, directly or indirectly, an enzyme with regulatable activity to said chromatin binding protein,b) transiently activating the regulatable enzymatic activity bound to the targeted chromatin andc) mapping the enzymatically-modified genomic sites introduced by the enzyme into the chromatin.
